AB0126 MULTIPARAMETER LINEBLOT IMMUNOASSAY FOR AUTOIMMUNE INFLAMMATORY MYOPATHIES: ADDITION OF cN-1A FOR THE DETECTION OF AUTOANTIBODIES ASSOCIATED WITH INCLUSION BODY MYOSITIS

چکیده
Background Autoantibodies against cN-1A (Mup44) are primarily used as a biomarker for inclusion body myositis (IBM), they also occur with lower prevalence in systemic lupus erythematosus (SLE) and Sjögren’s Syndrome (SjS). has been included parameter on the multiparameter lineblot Autoimmune Inflammatory Myopathies 16 Ag et (EUROIMMUN), allowing detection of 17 myositis-specific myositis-associated autoantibodies. Objectives The prevalences anti-cN-1A autoantibodies were determined different patient collectives healthy blood donors. Methods A immunoassay containing highly purified, recombinant expressed E.coli was incubated according to instructions. Sera 197 patients IBM, 48 collagenosis (systemic sclerosis, undifferentiated mixed tissue collagenosis), 52 polymyositis/dermatomyositis 75 SLE/SjS screened Additionally, 151 apparently donors well disease control collective comprising sera 20 thyroiditis, 26 neuromuscular disorders, 24 primary biliary cholangitis (PBC), 19 Wegener’s Disease, Coeliac disease, 2 autoimmune liver (ALD) analyzed. Signal intensities automatically evaluated using EUROLineScan software (EUROIMMUN). Results Prevalences 34.5% 10.4% collagenosis, 7.7% polymyositis/dermatomyositis, 13.3% SLE/SjS, 2% donor cohort 4.6% collective. Conclusion addition multiparametric now reduces risk underdiagnosing IBM inflammatory myopathies increases overall diagnostic sensitivity. [Inflammatory myopathies. Dermatomyositis, polymyositis, and inclusion body myositis].
Idiopathic inflammatory myopathies are a group of heterogeneous, acquired systemic diseases characterized by progressive symmetrical muscle weakness, elevated serum levels of muscle enzymes, electromyographic abnormalities, and inflammatory infiltrates on muscle biopsy. Characteristic histopathologic features allow classification of idiopathic inflammatory myopathies into polymyositis, dermatom...

OBJECTIVE To characterize serum autoantibody profiles of patients with idiopathic inflammatory myopathies (IIM) by searching for myositis-specific (MSA) and myositis-associated (MAA) antibodies with sensitive and specific laboratory tests. METHODS We tested the sera from 46 Caucasian patients diagnosed as affected with IIM at the Division of Rheumatology of Padova University (21 polimyositis,...

OBJECTIVE To compare familial inflammatory inclusion body myositis (IBM) with hereditary inclusion body myopathies and sporadic IBM. PATIENTS AND METHODS Clinical, biological, MRI, and histological data were analysed in two siblings with inflammatory IBM and compared with those of patients with sporadic and hereditary IBM. RESULTS Both patients had a clinical phenotype of sporadic IBM, whic...

Pathomechanisms of anti-cytosolic 5'-nucleotidase 1A autoantibodies in sporadic inclusion body myositis.
OBJECTIVE Sporadic inclusion body myositis (sIBM), an intractable progressive muscle disease, frequently occurs in older persons. sIBM pathogenesis may involve protein degradation dysfunction and immune abnormalities. Autoantibodies recognizing cytosolic 5'-nucleotidase 1A (cN1A) were found in plasma and serum from sIBM patients. However, whether anti-cN1A autoantibodies play a pathogenic role ...
